چکیده

The antiviral activity of oxidized spermine was compared with that of other aldehydes. Suspensions of vaccinia virus were incubated at 37 C with various concentrations of the aldehydes, and the infectivity of the viruses was determined by the plaque assay. Oxidized spermine at a concentration of 0.82 mM completely inactivated a suspension of vaccinia virus (1.4 x 10(8) plaque-forming units) after incubation at 37 C for 10 hr. Glutaraldehyde and formaldehyde were less active when compared on a molar basis, but acrolein resembled oxidized spermine in its antiviral activity. Because acrolein is produced from oxidized spermine at only 20 to 30% yield, it is unlikely that the biological activity of the latter is due to acrolein formed during the spontaneous degradation of oxidized spermine.

Evaluation of the effects of concentration, duration and temperature of incubation with Beta-Propiolactone on inactivation of cell-derived rabies virus

Introduction: During the production process of vaccines against viral diseases, virus inactivation plays a crucial role.  β-Propiolactone (BPL) is a compound that is frequently used for inactivation of viruses and production of the vaccines against viral diseases such as rabies. In this study, the effects of concentration of BPL as well as the duration and temperature of incubation with BPL on ...

The Tween 80 Toxicity in Chicken Embryos and Effect on the Kinetics of Newcastle Disease Virus Replication

Background: Non-ionic surfactant, Tween-80 (TW80) is commonly used for drug delivery due to its effect on the cell membrane permeation. The change in permeability can also increase viral infectivity in cells. This study was undertaken to improve upon Newcastle disease virus (NDV) titer cultivated with embryonic chicken eggs. Methods: The toxicity of TW80 was investigated against chicken embryo...

Infectivity and Capacity for D N A Replication of Vaccinia Virus Irradiated by 7-Rays

Purified vaccinia virus was irradiated by 7-rays under direct-effect conditions. The ability of the irradiated samples to form plaques (infectivity) and to induce viral DNA synthesis was determined. The radiosensitive volume of the viral unit causing infection (I.9 × IO-X7 cm2) is very small compared with the volume of the whole viral DNA ( ~ IO %). The inactivation of the DNA replication funct...

Enzymic inactivation of vaccinia virus with a bacterial protease.

pepsin (Dawson and McFarlane, 1948 ; Peter and Stoeckenius, 1954). For the purpose of finding out any enzyme to affect the surface structure of vaccinia virus under less radical conditions than with pepsin (pH 3.0), attempts were made to test various enzymes for the effect on the infectivity of the virus. Virus suspension was prepared from the homogenate of infected chorioallantoic membranes (C...

Effect of human leukocyte interferon on vaccinia-and herpes virus-infected cell cultures and monkey corneas.

Pretreatment of human fibroblast cultures with human leukocyte interferon (HIF, 1,000 IU/ml) resulted in a 24-h delay of virus replication after infection with vaccinia virus and herpes simplex virus type 1 and type 2. Additional HIF treatment 24 h after infection effectively lowered the maximum yield of viral infectivity. Equal results were obtained in simian cells with 3,000 IU of HIF per ml....
